ZKSCAN3 also known as Zinc finger protein with KRAB and SCAN domains 3 is a transcription factor with an approximate mass of 95 kDa. It contains zinc finger motifs that bind to DNA influencing the transcription of specific genes. ZKSCAN3 is expressed in various tissues with high levels noted in the kidney liver and lung. It plays a role in gene regulation by modulating transcriptional responses making it an important factor in cellular function.
ZKSCAN3 impacts cellular processes and functions as a transcriptional suppressor. ZKSCAN3 has been identified as part of certain protein complexes that interact with chromatin remodelers and co-regulators. This interaction helps in regulating gene expression critical for cell growth and development. Its activity influences various biological processes including cell cycle progression and apoptosis highlighting its role in maintaining cellular homeostasis.
ZKSCAN3 serves functions in the autophagy and Wnt signaling pathways. Within the autophagy pathway it negatively regulates autophagic activity impacting cellular homeostasis and survival under stress conditions. ZKSCAN3 is related to proteins like LC3 and p62 which are significant in this pathway. Its interaction within the Wnt signaling pathway contributes to the regulation of gene expression related to proliferation and differentiation with beta-catenin as a notable interacting protein.
ZKSCAN3 shows implications in cancer and neurodegenerative diseases. In cancers its overexpression correlates with tumor progression and poor prognosis often through interaction with proteins such as MYC and mTOR which promote cancer cell survival. Regarding neurodegenerative disorders alterations in ZKSCAN3 expression influence pathways linked with neuronal survival and autophagy affecting disease mechanisms in disorders such as Alzheimer’s disease. These connections make ZKSCAN3 an interesting target for therapeutic research.
