Supplementary information

ZNF408 also known as Zinc Finger Protein 408 is a protein with a mass of approximately 95 kDa. It belongs to the family of zinc finger proteins which play essential roles in DNA binding and transcription regulation. ZNF408 locates primarily in the nucleus of cells where it binds specific DNA sequences to influence the transcriptional activity. It expresses in various tissues with notable levels detected in the brain and heart suggesting a role in the functioning of these organs.
Biological function summary

This protein participates in the regulation of gene expression important for maintaining cellular homeostasis and function. ZNF408 is known to interact with DNA and potentially with other transcription factors forming a complex that modulates transcription. Through its zinc finger domains ZNF408 recognizes particular gene promoters influencing the transcription of genes related to cellular growth and development processes.

Pathways

Four pertinent biological interactions involve ZNF408's influence on the Wnt signaling pathway and possibly the Hedgehog signaling pathway. These pathways are important for proper embryonic development and maintaining adult tissue homeostasis. In these cascades ZNF408 interacts with other transcriptional regulators such as β-catenin in the Wnt pathway highlighting its role in modulating transcription in response to cellular signals.

ZNF408's function links to retinal diseases and certain cancers. Mutations or dysregulations in this protein may contribute to retinal degeneration potentially through its interaction with proteins like RPGRIP1. Additionally aberrant activity or expression of ZNF408 associates with cancer progression showing interactions with tumor suppressor proteins or oncogenes. Understanding ZNF408's detailed role may aid in designing targeted therapies for these conditions.
