Supplementary information

ZNF45 also known as Zinc Finger Protein 45 belongs to the zinc finger protein family. It acts as a transcription factor and possesses zinc finger domains which helps in DNA binding and regulation of gene expression. The estimated mass of ZNF45 is about 54 kDa. Analysis shows an expression of ZNF45 in various tissues with higher levels observed in specific cell types particularly in the brain and liver. The protein localizes in the nucleus where it directly associates with DNA to exert its functional activities.
Biological function summary

Zinc Finger Protein 45 regulates the expression of genes involved in cell growth and apoptosis. ZNF45 forms part of a larger protein complex which includes other zinc finger proteins and co-factors that modulate its activity. These interactions enable ZNF45 to control various cell processes such as proliferation and differentiation. Through its role in transcription regulation ZNF45 affects the overall cellular homeostasis which helps in maintaining normal cellular functions and responses to environmental signals.

Pathways

The Zinc Finger Protein 45 influences the p53 signaling pathway which plays a critical role in cell cycle regulation and apoptosis. As a transcriptional regulator ZNF45 interacts with other proteins such as MDM2 to modulate the activity of the p53 pathway. Additionally ZNF45 is part of the Wnt signaling pathway where it associates with proteins like β-catenin to regulate the expression of target genes involved in cell cycle progression and oncogenesis. These pathways are essential for maintaining cellular integrity and preventing uncontrolled cell growth.

The altered expression of Zinc Finger Protein 45 associates with several cancers including liver cancer and brain tumors. In liver cancer ZNF45 interacts with proteins such as p53 whose pathway dysregulation can contribute to tumorigenesis. Similarly in brain tumors aberrant expression of ZNF45 may influence pathways involving β-catenin promoting cancer cell survival and proliferation. Understanding the role of ZNF45 in these diseases may provide new insights into potential therapeutic targets for cancer treatment.
