Supplementary information

ZNF638 also known as ZNF638 zinc finger protein is a nuclear protein weighing approximately 197 kDa. This protein features multiple zinc finger domains which are characteristic structural motifs that enable binding to nucleic acids. Researchers find ZNF638 expressed across various tissues with high levels in the brain and testis. It functions as a regulator of gene transcription influencing the expression of numerous genes involved in cell differentiation and proliferation.
Biological function summary

ZNF638 plays an important role in coordinating the cellular processes necessary for adipogenesis and neural development. It acts as a transcription factor and shows interaction with several other transcriptional regulators. ZNF638 also forms part of larger protein complexes involved in the modulation of chromatin structure which is important for its role in transcription regulation. By facilitating chromatin remodeling ZNF638 allows access to gene targets necessary for specific cell lineages development.

Pathways

The functional activities of ZNF638 influence the PPAR signaling pathway which is essential for the regulation of adipocyte differentiation. The protein interacts with PPARγ an important regulator within this pathway as well as with other PPAR-associated proteins such as C/EBPα. Additionally ZNF638 involvement is evident in the regulation of the Wnt signaling pathway where it interacts with LEF1/T-cell factor to modulate the transcriptional activity involved in cell fate determination during embryonic development and tissue homeostasis.

Abnormal expression or function of ZNF638 has been linked to metabolic disorders like obesity and potentially schizophrenia. Dysregulation in adipogenesis due to altered ZNF638 activity can contribute to obesity through its interaction with proteins like PPARγ. In the context of schizophrenia variations in the ZNF638 gene expression or activity might affect neural development interacting with proteins like DISC1 a protein associated with cognitive functioning and psychiatric disorders.

Target data

Transcription factor that binds to cytidine clusters in double-stranded DNA (PubMed : 30487602, PubMed : 8647861). Plays a key role in the silencing of unintegrated retroviral DNA : some part of the retroviral DNA formed immediately after infection remains unintegrated in the host genome and is transcriptionally repressed (PubMed : 30487602). Mediates transcriptional repression of unintegrated viral DNA by specifically binding to the cytidine clusters of retroviral DNA and mediating the recruitment of chromatin silencers, such as the HUSH complex, SETDB1 and the histone deacetylases HDAC1 and HDAC4 (PubMed : 30487602). Acts as an early regulator of adipogenesis by acting as a transcription cofactor of CEBPs (CEBPA, CEBPD and/or CEBPG), controlling the expression of PPARG and probably of other proadipogenic genes, such as SREBF1 (By similarity). May also regulate alternative splicing of target genes during adipogenesis (By similarity).
