Supplementary information

ZNF750 also known as zinc finger protein 750 is a transcription factor with a molecular weight of approximately 80 kDa. It contains zinc finger motifs that allow it to bind DNA and regulate gene expression. This protein is primarily expressed in the epidermis where it plays a critical role in cellular differentiation. ZNF750 acts by modulating the expression of genes involved in the maintenance and function of the skin barrier.
Biological function summary

This transcription factor influences the differentiation of keratinocytes the predominant cell type in the epidermis. ZNF750 controls the terminal differentiation programs by inducing the expression of differentiation-specific genes. It functions in part of a larger transcriptional complex interacting with other transcription factors and co-regulators to execute its functions. These interactions dictate the precise genetic output necessary for maintaining skin integrity.

Pathways

ZNF750 plays a part in the epidermal differentiation complex pathway. This pathway coordinates the transition of keratinocytes from proliferative basal cells to differentiated cells. The activity of ZNF750 closely links with other proteins like KLF4 and GRHL3 which are also important in epidermal regulation. Together these proteins form a network that supports skin development and repair processes.

Mutations or dysfunctions in ZNF750 have associations with skin-related conditions such as psoriasis and seborrheic dermatitis. Aberrant expression or mutations can disrupt normal keratinocyte differentiation leading to hyperproliferative skin disorders. ZNF750's activity connects with other factors including c-MYC which is involved in cellular proliferation highlighting its role in maintaining the balance between cell proliferation and differentiation in the epidermis.

Target data

Transcription factor involved in epidermis differentiation. Required for terminal epidermal differentiation : acts downstream of p63/TP63 and activates expression of late epidermal differentiation genes. Specifically binds to the promoter of KLF4 and promotes its expression.
