Supplementary information

ZNHIT2 also known as Zinc Finger HIT-Type Containing 2 is a protein that plays a role in transcription regulation by modulating the activity of other proteins. With a molecular mass of approximately 31 kDa this protein is characterized by the presence of a zinc-finger HIT domain. ZNHIT2 is expressed in various tissues including the brain testis and heart indicating its involvement in diverse physiological processes. Its expression pattern suggests a significant role in cellular activities affecting many tissue types.
Biological function summary

ZNHIT2 participates in the assembly and function of essential ribonucleoprotein complexes. It acts as a facilitator in the modification of these complexes ensuring proper ribosome maturation and function. ZNHIT2 is part of larger multi-subunit complexes that include proteins from the U3 snoRNP complex impacting the processing of precursor rRNA. Its presence in these complexes highlights its involvement in maintaining the efficiency of cellular protein synthesis mechanisms.

Pathways

ZNHIT2 plays a role in the ribosome biogenesis pathway. It directly interacts with other proteins like NOLC1 and BOP1 which are important for the production of functional ribosomes. These interactions ensure the proper progression and regulation of ribosomal assembly and function making ZNHIT2 an important player in the protein synthesis pathway. By coordinating with these proteins ZNHIT2 helps maintain cellular homeostasis and protein production.

Alterations in ZNHIT2 expression have been implicated in neurodegenerative diseases and certain cancers. ZNHIT2's interaction with proteins such as NOP56 and DDX21 links it to disorders like spinocerebellar ataxias (SCAs) where ribosome biogenesis is disrupted. Additionally in some cancers ZNHIT2 expression levels may affect tumor progression due to its influence on cellular growth mechanisms. Understanding how ZNHIT2 functions in these contexts can provide insights into potential therapeutic strategies.

Target data

May act as a bridging factor mediating the interaction between the R2TP/Prefoldin-like (R2TP/PFDL) complex and U5 small nuclear ribonucleoprotein (U5 snRNP) (PubMed : 28561026). Required for the interaction of R2TP complex subunit RPAP3 and prefoldin-like subunit URI1 with U5 snRNP proteins EFTUD2 and PRPF8 (PubMed : 28561026). May play a role in regulating the composition of the U5 snRNP complex (PubMed : 28561026).
