Znt6 also known as SLC30A6 is a zinc transporter protein involved in the homeostatic regulation of zinc ions. The protein has a molecular weight of approximately 45 kDa. Znt6 facilitates the transport of zinc ions across cellular membranes particularly into the Golgi apparatus and vesicles. This transporter is widely expressed in many tissues but its expression is notably high in the pancreas liver and brain.
Biological function summary

Znt6 plays a role in maintaining cellular zinc homeostasis. It is part of the cation diffusion facilitator (CDF) family involved in transporting metal ions within cells. Znt6 contributes to the sequestration of zinc into intracellular compartments which is important for zinc-dependent processes and enzyme functions. This protein is not known to be part of any larger complexes but works in coordination with other zinc transporters to balance intracellular zinc levels efficiently.

Pathways

Znt6 participates in cellular processes such as zinc ion transmembrane transport and metal ion homeostasis. It is linked to the regulation of pathways such as the MAPK signaling pathway which is influenced by zinc levels and the cellular response to oxidative stress. Proteins such as Znt7 another zinc transporter also interacts with related pathways ensuring the proper function of metalloenzymes and zinc-dependent cellular activities.

Znt6 has been associated with neurodegenerative diseases like Alzheimer's disease and metabolic disorders like diabetes. Dysregulation of zinc homeostasis in which Znt6 plays an important part can disrupt neuronal signaling and metal ion regulation. In Alzheimer's disease altered zinc transport involving Znt6 can affect amyloid-beta metabolism. In diabetes improper zinc regulation is linked to insulin granule formation and secretion. Proteins linked to zinc transporters like zinc transporter 8 (Znt8) also exhibit related dysfunctions in diabetes highlighting the interconnected role of Znt6 in disease states.

Has probably no intrinsic transporter activity but together with SLC30A5 forms a functional zinc ion : proton antiporter heterodimer, mediating zinc entry into the lumen of organelles along the secretory pathway (PubMed : 15994300, PubMed : 19366695, PubMed : 19759014).
