Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from A-549 cells was prepared using NETN lysis buffer. ab276131 was used for IP at 12 μl per reaction (Lane 1). TJP1/ZO-1 was also immunoprecipitated by rabbit anti-TJP1/ZO-1 antibody. For blotting immunoprecipitated ab276131 was used at 1/1000 dilution. Contol IgG shown in Lane 2.

Western blot : Anti-TJP1 antibody [BLR092G] (ab276131) staining at 1/1000 dilution, shown in green; Mouse anti-Alpha Tubulin [DM1A] (ab7291) loading control staining at 1/20000 dilution, shown in magenta. In Western blot, ab276131 was shown to bind specifically to TJP1. A band was observed at 195 kDa in wild-type HeLa cell lysates with no signal observed at this size in TJP1 knockout cell line. There is a close non-specific band at 190 kDa. We recommend running a low percentage gel to separate this from the band of interest. To generate this image, wild-type and TJP1 knockout HeLa cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L 800CW and Goat anti-Mouse IgG H&L 680RD at 1/20000 dilution.

The ZO1 tight junction protein also known as TJP1 is a critical component of tight junctions in epithelial and endothelial cells. It is a 225 kDa protein that plays a mechanical role in stabilizing cell-cell junctions by linking the actin cytoskeleton to tight junction membrane proteins. ZO1 is expressed in various tissues including the liver kidney and brain where it is essential for maintaining cell polarity and permeability barriers. It acts as a scaffold facilitating the organization of other tight junction proteins.

TJP1, TJP2, and TJP3 are closely related scaffolding proteins that link tight junction (TJ) transmembrane proteins such as claudins, junctional adhesion molecules, and occludin to the actin cytoskeleton (PubMed : 7798316, PubMed : 9792688). Forms a multistranded TJP1/ZO1 condensate which elongates to form a tight junction belt, the belt is anchored at the apical cell membrane via interaction with PATJ (By similarity). The tight junction acts to limit movement of substances through the paracellular space and as a boundary between the compositionally distinct apical and basolateral plasma membrane domains of epithelial and endothelial cells. Necessary for lumenogenesis, and particularly efficient epithelial polarization and barrier formation (By similarity). Plays a role in the regulation of cell migration by targeting CDC42BPB to the leading edge of migrating cells (PubMed : 21240187). Plays an important role in podosome formation and associated function, thus regulating cell adhesion and matrix remodeling (PubMed : 20930113). With TJP2 and TJP3, participates in the junctional retention and stability of the transcription factor DBPA, but is not involved in its shuttling to the nucleus (By similarity). May play a role in mediating cell morphology changes during ameloblast differentiation via its role in tight junctions (By similarity).
