ZPI also known as Protein Z-dependent protease inhibitor is a serine protease inhibitor with a mass of approximately 72 kDa. It inhibits factor Xa in a process that requires Protein Z and calcium ions. ZPI is mainly expressed in the liver and its presence has been noted in plasma. Structurally ZPI acts by forming a complex with factor Xa in the presence of protein Z leading to the inactivation of the coagulation pathway's enzymatic activity effectively reducing clot formation under physiological conditions.
ZPI plays a significant role in the regulation of hemostasis controlling blood clot formation by inhibiting excess factor Xa activity. This inhibition prevents unnecessary clotting maintaining fluidity and preventing vascular occlusion. ZPI is particularly important in the context of the coagulation cascade where it forms part of a complex with Protein Z and calcium ions offering a fine balance to the procoagulant processes mediated by thrombin and factor Xa.
The actions of ZPI largely influence the intrinsic and extrinsic coagulation pathways. Its primary function is to regulate the activity of factor Xa significantly impacting the common pathway of coagulation. Through these pathways ZPI is closely related to proteins like factor VIIa and factor IXa participating in a delicate interplay necessary for maintaining hemostatic balance and preventing thrombosis or hemorrhage.
ZPI's functional integrity is linked to conditions such as thrombophilia where an inability to effectively inhibit factor Xa can lead to increased thrombotic events. Furthermore deficiencies or malfunctions of ZPI have been associated with a heightened risk of venous thromboembolism (VTE). Altered interactions between ZPI and protein Z or factor Xa can contribute to these conditions highlighting the importance of balanced protease inhibitor activity in maintaining vascular health.
