ZWILCH was immunoprecipitated from 1mg of HeLa (Human epithelial cells from cervix adenocarcinoma) whole cell lysate with ab202898 at 1/50 dilution. Western blot was performed from the immunoprecipitate using ab202898 at 1/1000 dilution. Anti-Rabbit IgG (HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/1500 dilution.
Lane 1 : HeLa whole cell lysate 10ug (Input). Lane 2 : ab202898 IP in HeLa whole cell lysate. Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab202898 in HeLa whole cell lysate.
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
Exposure time : 10 seconds.

Biological function summary
ZWILCH is fundamental in ensuring accurate chromosome alignment and segregation during cell division. It plays a part as a component of the RZZ complex. The RZZ complex localizes to kinetochores which are critical for connecting spindle microtubules to chromosomes promoting bioriented chromosome alignment. Moreover ZWILCH contributes to the spindle assembly checkpoint a safety mechanism that prevents chromosome mis-segregation by delaying progress to anaphase until all chromosomes are correctly attached.
Pathways
ZWILCH plays integral roles in the mitotic spindle checkpoint and chromosomal stability pathways. Within these pathways ZWILCH associates closely with spindle assembly checkpoint proteins such as MAD2 and BUBR1 which monitor kinetochore-microtubule attachment. ZWILCH through its role in the RZZ complex communicates tension status between sister chromatids ensuring proper cell-cycle progression and genomic integrity.
