In vivo antibodies for neutrophil depletion
The hybridoma clone, anti-Ly6G [1A8] rat IgG2a, is widely used for neutrophil depletion. However, due to the rapid self-renewing nature of neutrophils, it is only effective for 48 hours, meaning experiments that use this antibody require a lot of hands-on time. To reduce repetitive injections, we murinized the existing Ly6G [1A8] clone, resulting in increasing its half-life and reducing anti-rat antibody production in treated mice, which otherwise compromised efficacy in long-term studies.
