Neutrophils are indispensable to human health due to their pivotal role in the immune system, acting as the body's first line of defense against bacterial and fungal infections. These white blood cells are highly motile and can rapidly respond to sites of infection or injury. Upon arrival, they engulf and destroy invading pathogens through phagocytosis and release toxic substances such as reactive oxygen species and antimicrobial peptides. Neutrophils also form neutrophil extracellular traps (NETs), which ensnare and kill microbes, preventing their spread and facilitating their clearance. By rapidly containing and eliminating pathogens, neutrophils help to prevent infections from becoming severe or widespread, thus playing a key role in maintaining health.
In addition to their antimicrobial functions, neutrophils are integral to the regulation of inflammation and tissue repair. They release cytokines and chemokines that recruit other immune cells to the site of infection, orchestrating a coordinated immune response. Furthermore, neutrophils aid in clearing debris and dead cells, promoting tissue regeneration and wound healing. However, their dysfunction can lead to significant health issues. Conditions such as neutropenia, characterized by low neutrophil counts, increase susceptibility to infections, while overactive neutrophils can contribute to tissue damage in autoimmune diseases like rheumatoid arthritis. In severe infections, an excessive neutrophil response can result in systemic inflammation and organ damage, as seen in sepsis. Thus, the proper function of neutrophils is important for balancing effective immune defense with controlled inflammation, underscoring their importance in both health and disease.
In vivo antibodies for neutrophil depletion
Neutrophil depletion, the significant reduction in the number of neutrophils, is a critical area of research for understanding human diseases, as these white blood cells are pivotal in the immune response against infections. Neutrophil depletion can be induced by factors such as chemotherapy, radiation, autoimmune diseases, or severe infections, leading to increased susceptibility to bacterial and fungal pathogens. The hybridoma clone, anti-Ly6G [1A8] rat IgG2a, is widely used in the neutrophil research community. However, due to the rapid self-renewing nature of neutrophils, it is only effective for 48 hours, meaning experiments that use this antibody require a lot of hands-on time.
To reduce repetitive injections, we murinized the existing Ly6G [1A8] clone, resulting in increasing its half-life and reducing anti-rat antibody production in treated mice, which otherwise compromised efficacy in long-term studies.
We successfully converted the rat IgG2a hybridoma anti-Ly6G [1A8] to a recombinant antibody and murinized it, creating two new chimeric antibodies: mouse IgG2a anti-Ly6G [1A8] and mouse IgG2c anti-Ly6G [1A8]. Having different IgG2 types allows you to use it in different mice backgrounds.
