Tools for neutrophil depletion and analysis

IgorE Abcam LN2 Aliquot

Understanding the intricacies of neutrophil activity can help advance your research on immune response, inflammation, and disease progression, from infection control to autoimmune disorders. However, we understand the challenges of studying neutrophils. That is why we are dedicated to providing you with reliable, high-quality tools to explore neutrophils in depth. Unlock new insights into immune function with our range of neutrophil depletion kits, specific antibodies, and advanced detection assays.

Neutrophils are indispensable to human health due to their pivotal role in the immune system, acting as the body's first line of defense against bacterial and fungal infections. These white blood cells are highly motile and can rapidly respond to sites of infection or injury. Upon arrival, they engulf and destroy invading pathogens through phagocytosis and release toxic substances such as reactive oxygen species and antimicrobial peptides. Neutrophils also form neutrophil extracellular traps (NETs), which ensnare and kill microbes, preventing their spread and facilitating their clearance. By rapidly containing and eliminating pathogens, neutrophils help to prevent infections from becoming severe or widespread, thus playing a key role in maintaining health.

In addition to their antimicrobial functions, neutrophils are integral to the regulation of inflammation and tissue repair. They release cytokines and chemokines that recruit other immune cells to the site of infection, orchestrating a coordinated immune response. Furthermore, neutrophils aid in clearing debris and dead cells, promoting tissue regeneration and wound healing. However, their dysfunction can lead to significant health issues. Conditions such as neutropenia, characterized by low neutrophil counts, increase susceptibility to infections, while overactive neutrophils can contribute to tissue damage in autoimmune diseases like rheumatoid arthritis. In severe infections, an excessive neutrophil response can result in systemic inflammation and organ damage, as seen in sepsis. Thus, the proper function of neutrophils is important for balancing effective immune defense with controlled inflammation, underscoring their importance in both health and disease.

In vivo antibodies for neutrophil depletion

Neutrophil depletion, the significant reduction in the number of neutrophils, is a critical area of research for understanding human diseases, as these white blood cells are pivotal in the immune response against infections. Neutrophil depletion can be induced by factors such as chemotherapy, radiation, autoimmune diseases, or severe infections, leading to increased susceptibility to bacterial and fungal pathogens. Studying neutrophil depletion can help you uncover the mechanisms of immune system failure and the body's vulnerability to infections, providing insights into the development of targeted therapies and preventative measures.

The hybridoma clone, anti-Ly6G [1A8] rat IgG2a, is widely used in the neutrophil research community. However, due to the rapid self-renewing nature of neutrophils, it is only effective for 48 hours, meaning experiments that use this antibody require a lot of hands-on time.
To reduce repetitive injections, we murinized the existing Ly6G [1A8] clone, resulting in increasing its half-life and reducing anti-rat antibody production in treated mice, which otherwise compromised efficacy in long-term studies.

We successfully converted the rat IgG2a hybridoma anti-Ly6G [1A8] to a recombinant antibody and murinized it, creating two new chimeric antibodies: mouse IgG2a anti-Ly6G [1A8] and mouse IgG2c anti-Ly6G [1A8]. Having different IgG2 types allows you to use it in different mice backgrounds.

Conjugation kits

Conjugation of primary antibodies can result in antibody loss and consume significant laboratory time. Our cutting-edge Lightning-Link® labeling technology facilitates the direct labeling of antibodies or proteins without the need for purification, offering a simple alternative to conventional conjugation techniques and improving the efficiency of the research and development process.

Our Lightning-Link® conjugation kits are easy to use and allow you to conjugate antibodies in three simple steps to more than 45 labels, including fluorescent proteins commonly used in flow cytometry (Alexa Fluor® dyes, tandem dyes, oligonucleotides, metal isotopes) as well as other labels such as gold nanoparticles, and DNA barcodes, and only require 30 seconds of hands-on time so you can focus on your research. Our Lightning-Link® conjugation kits are ready to be used with our extensive range of 13,000 conjugation-ready carrier-free antibodies, which are free from glycerol, BSA, and sodium azide to reduce antibody loss and maximize conjugation efficiency, giving you the ultimate flexibility to design and complete your experiment.

If your antibody does not meet these buffer requirements, you can easily purify or concentrate it using our range of purification and concentration kits.
