Recombinant E. coli Multi Tag protein is a protein, expressed in Escherichia coli and suitable for WB.

Constituents: 10% Glycerol (glycerin, glycerine), 2% Sodium lauryl sulfate, 0.76% Tris, 0.01% 3,3-bis(3,5-dibromo-4-hydroxyphenyl)-3H-2,1λ⁶-benzoxathiole-1,1-dione
This protein lysate contains the commonly used epitope tags: HA, c-myc, Glu-Glu, GST, 6-His, HSV, T7, V5, VSV, E-tag, S-tag, FLAG (TM). These proteins were fused in the following order, GST, T7, HSV, c-myc, VSV, Glu-Glu, V5, E-tag, DYKDDDDK, S-tag and HA, 6xHis.
This product is a bacterial lysate provided in1 x reduced SDS-PAGE loading buffer. It is effective as a positive control for epitope tag antibodies in immunoblotting.
For Western blotting, the suggested use of this reagent is 0.5 - 2.0 μl. It is recommended that the reagent be titrated for optimal performance for each application.

Anti-V5 tag antibody [SV5-P-K] staining at 1/1000 dilution, shown in black. In Western blot, Anti-V5 tag antibody [SV5-P-K] ab206566 was shown to bind specifically to V5 tag. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween$®$ 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times before development with Optiblot (ECL reagent Anti-PKC delta (phospho S299) antibody [EPNCI119] ab133456) and imaged with 3 minutes exposure time. Secondary antibodies used were HRP conjugated Goat anti-Rabbit (H+L) at 1/50000 dilution.
