Biological function summary
The ALKTRK fusion protein participates in abnormal cellular signaling that can lead to uncontrolled cell growth. Normally both ALK and TRK proteins function in neuronal development and signaling but when fused the resulting protein loses normal regulatory control. This fusion protein is not generally part of a larger complex but acts independently to drive oncogenic processes. ALK a receptor tyrosine kinase activates pathways that promote cell proliferation and survival mimicked in the fusion form with TRK proteins.
Pathways
The ALKTRK fusion protein engages primarily in the MAPK/ERK and PI3K/AKT pathways which are critical for cell proliferation and survival. In its normal non-fused state ALK interacts in pathways with proteins like IRS1 and GRB2 while TRK partners with proteins such as SHC and PLC-γ. The fusion form activates similar downstream signaling pathways bypassing normal cellular controls and frequently leading to malignancy.
