The ALKTRK fused gene product known as a fusion protein combines parts of the anaplastic lymphoma kinase (ALK) and neurotrophic receptor tyrosine kinase (TRK) genes. The fusion protein has a molecular mass usually in the range of 75–200 kDa dependent on the exact fusion partners and the number of amino acids in the resulting protein. Researchers identify these proteins mainly in cancer cells where they play a role in oncogenic signaling. ALK typically shows expression in the brain and nervous system while TRK proteins express in the nervous system and various other tissues during development.
Biological function summary

The ALKTRK fusion protein participates in abnormal cellular signaling that can lead to uncontrolled cell growth. Normally both ALK and TRK proteins function in neuronal development and signaling but when fused the resulting protein loses normal regulatory control. This fusion protein is not generally part of a larger complex but acts independently to drive oncogenic processes. ALK a receptor tyrosine kinase activates pathways that promote cell proliferation and survival mimicked in the fusion form with TRK proteins.

The ALKTRK fused gene product associates closely with certain cancers particularly non-small cell lung cancer and some pediatric malignancies. This fusion protein plays a significant role in these diseases by driving oncogenic transformation and progression. Specifically TRK fusions have been implicated in various solid tumors while ALK fusions are prominently observed in anaplastic large cell lymphoma. Targeting the ALKTRK fusion protein with specific inhibitors can lead to effective therapies for these cancers.
