Biological function summary
B2M interacts with the neonatal Fc receptor and is involved in the regulation of IgG antibody levels by extending their half-life. As part of the FcRn complex B2M also aids in the transport of IgG across epithelial barriers such as the intestinal lining in neonatal mammals. This complex plays a significant role in maintaining the homeostasis of IgG antibodies therefore impacting adaptive immunity.
Pathways
B2M participates in antigen processing and presentation. It forms a complex with FCGRT which is critical in the FcRn pathway responsible for the recycling and transcytosis of immunoglobulins. This interaction preserves IgG from lysosomal degradation. Additionally B2M interacts with other proteins like albumin through related pathways highlighting its involvement in protein recycling and homeostasis.

General info
Function
Component of the class I major histocompatibility complex (MHC). Involved in the presentation of peptide antigens to the immune system. Exogenously applied M.tuberculosis EsxA or EsxA-EsxB (or EsxA expressed in host) binds B2M and decreases its export to the cell surface (total protein levels do not change), probably leading to defects in class I antigen presentation (PubMed : 25356553).
Sequence similarities
Belongs to the beta-2-microglobulin family.
Post-translational modifications
Glycation of Ile-21 is observed in long-term hemodialysis patients.
