Biological function summary

The protein exerts its function by promoting apoptosis. Bmf binds directly to pro-survival Bcl-2 family members such as Bcl-X_L and Bcl-2 displacing pro-apoptotic proteins like Bax or Bak. This action facilitates the release of cytochrome c from mitochondria triggering caspase activation and cell death. Bmf can function independently but also works within larger complexes affecting processes related to cellular homeostasis.

Activity summary

Bmf also known as Bcl-2 modifying factor is important in apoptosis regulation a process of programmed cell death. It acts as a pro-apoptotic protein from the Bcl-2 family. The Bmf protein has a molecular weight of approximately 21 kDa. It is mainly expressed in various epithelial tissues and abundant in hematopoietic cells. Bmf can interact with the dynein light chain at the cytoskeleton which affects its localization and function.

Pathways

Bmf operates prominently within the intrinsic apoptotic pathway where it modulates mitochondrial outer membrane permeability. Its action is closely associated with Bax and Bak proteins facilitating their oligomerization. Additionally Bmf integrates into stress-induced signaling pathways responding to signals like oxidative stress or deprivation of survival factors. Through these connections Bmf influences cell fate and stress responses often cross-talking with the JNK signaling pathway.

Associated diseases and disorders

Bmf has implications in several types of cancer and neurodegenerative conditions. Overexpression or dysregulation of Bmf can contribute to tumor development due to impaired apoptosis. Specific cancers like lymphomas show altered Bmf levels linking it to pathways with proteins such as Bcl-2 and Bax.
