Supplementary information

C-4 Methylsterol Oxidase also known as SC4MOL is an important enzyme involved in cholesterol biosynthesis. This enzyme is responsible for the demethylation of 44-dimethylsterols in the cholesterol synthesis process. It is a protein with a molecular mass of approximately 47 kDa. SC4MOL is widely expressed across various tissues including the liver where it has a significant role in lipid metabolism.
Biological function summary

The role of C-4 Methylsterol Oxidase is important for the efficient production of cholesterol which is a component of cell membranes and precursor for steroid hormones. This enzyme belongs to the sterol C4-methyl oxidase complex contributing to the fine-tuning of sterol intermediates during their conversion to cholesterol. Its activity ensures the reduction of certain sterol precursors and the stability of membrane fluidity across diverse physiological conditions.

Pathways

The enzyme is involved in the Kandutsch-Russell pathway and the Bloch pathway both of which are essential for cholesterol biosynthesis. These pathways manage the flow of sterol intermediates toward cholesterol while maintaining cellular homeostasis. SC4MOL interacts with other important enzymes like HMG-CoA reductase and lanosterol synthase playing a critical role in these metabolic processes.

SC4MOL has links to conditions such as Conradi–Hünermann syndrome and multiple sclerosis. Mutations or deficiencies in SC4MOL may result in abnormal sterol accumulation leading to these disorders. In Conradi–Hünermann syndrome the pathway disruption caused by SC4MOL impacts the processing of specific sterols while in multiple sclerosis cholesterol biosynthesis abnormalities can influence myelin sheath integrity associating the enzyme with this neurological condition through its interaction with other cholesterol-related proteins.

General info

Function

Catalyzes the three-step monooxygenation required for the demethylation of 4,4-dimethyl and 4alpha-methylsterols, which can be subsequently metabolized to cholesterol (PubMed : 21285510, PubMed : 23583456, PubMed : 26114596, PubMed : 28673550, PubMed : 36958722). Also involved in drug metabolism, as it can metabolize eldecalcitol (ED-71 or 1alpha,25-dihydroxy-2beta-(3-hydroxypropoxy)-cholecalciferol), a second-generation vitamin D analog, into 1alpha,2beta,25-trihydroxy vitamin D3; this reaction occurs via enzymatic hydroxylation and spontaneous O-dehydroxypropylation (PubMed : 26038696).

Sequence similarities

Belongs to the sterol desaturase family.

Post-translational modifications

Ubiquitinated by MARCHF6, leading to proteasomal degradation.
