The CCNO gene encodes the Cyclin O protein which plays a role in cell cycle regulation particularly in the G2/M transition phase. Cyclin O a member of the cyclin protein family has a molecular mass of approximately 43 kDa. It is expressed in various tissues with higher expression observed in respiratory epithelial cells. Cyclin O interacts with cyclin-dependent kinases (CDKs) which are key regulators of cell cycle progression. This interaction is essential for maintaining proper cell division and genomic stability.
Cyclin O contributes to ciliogenesis the process by which cells build cilia. Cyclin O is part of a multiprotein complex that includes components necessary for the formation and function of motile cilia. Cilia play an important role in cellular processes such as signaling and movement of fluids across cell surfaces. Disruption in the cyclin O function can result in compromised ciliary function. This can impact various physiological processes like mucus clearance in respiratory pathways and fluid movement in brain ventricles.
Cyclin O is involved in the ciliogenesis pathway. It interacts with several other proteins like CDC28 and those involved in regulatory pathways of cell division ensuring that cilia are properly assembled and functional. Cyclin O's involvement in the proper assembly and maintenance of cilia illustrates its regulatory function within the Wnt signaling pathway which is a pathway critical for cell proliferation differentiation and polarity.
Cyclin O is associated with primary ciliary dyskinesia (PCD) a genetic disorder that affects the structure and function of cilia leading to chronic respiratory tract infections and problems in the reproductive system. Mutations in the CCNO gene have also been implicated in the development of certain ciliopathies disorders characterized by abnormalities in ciliary function. Cyclin O's relationship with proteins such as dynein is particularly noted in conditions where motility of cilia is affected linking it to the pathogenesis of these ciliopathy-related conditions.
