Clathrin light chain B often known as CLTB is a component of the clathrin triskelion a structure important in endocytosis. It has a molecular mass of about 25 kDa and is ubiquitously expressed in various tissues with abundant presence in neuronal and non-neuronal cells. The clathrin triskelion consists of three heavy chains and three light chains including the clathrin light chains A and B forming a scaffold that facilitates the formation of clathrin-coated vesicles.
Biological function summary

Clathrin light chain B plays an essential role in the assembly of clathrin-coated vesicles a process fundamental for receptor-mediated endocytosis. It is part of the clathrin complex interacting with adaptor protein complexes and helping them to coat and stabilize formed vesicles. This enables the transport of molecules into the cell recycling of receptors and intracellular signaling processes. The light chains are important for regulating the lattice structure of clathrin influencing the vesicle size and curvature.

Pathways

Clathrin light chain B is integral in endocytic and vesicular trafficking pathways. It is associated with the AP2 adaptor complex which is important for selecting cargo for clathrin-mediated endocytosis. Additionally the protein intersects with the regulated secretory pathway interacting with proteins such as dynamin and amphiphysin which assist in membrane scission during vesicle formation.

Dysregulation of clathrin light chain B appears linked to neurodegenerative disorders like Alzheimer's disease and Huntington's disease. Abnormalities in clathrin-mediated endocytosis disrupt neuronal homeostasis. In these conditions misprocessing of amyloid precursor protein involves clathrin complexes including CLTB signifying its role. Collaborators in this process include proteins like presenilin-1 and beta-secretase which are involved in the generation of amyloid-beta peptides contributing to plaque formation.
