Membranes were blocked in 3 % milk in TBS-T (0.1 % Tween®) before incubation with ab271180 and ab18184 (Mouse monoclonal [HIS.H8] to 6X His tag®) overnight at 4 °C at a 1 in 1000 dilution of both antibodies. Blots were incubated with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) secondary antibodies at 1 in 20000 dilution for 1 h at room temperature before imaging.

The nucleocapsid protein of SARS-CoV-2 also known as N protein or COVID-19 nucleocapsid protein is important for the viral replication and packaging processes. It has a mass of approximately 43-46 kDa and is expressed in the cytoplasm of infected host cells. This protein is highly conserved among coronaviruses and plays a significant role in the viral lifecycle by binding to the viral RNA genome thereby facilitating its encapsulation within the virion.
