Function

Responsible for the deiodination of T4 (3,5,3',5'-tetraiodothyronine) into T3 (3,5,3'-triiodothyronine) and of T3 into T2 (3,3'-diiodothyronine). Plays a role in providing a source of plasma T3 by deiodination of T4 in peripheral tissues such as liver and kidney.

Activity summary

Type I iodothyronine deiodinase also known as DIO1 plays an important role in thyroid hormone metabolism by catalyzing the conversion of thyroxine (T4) to the more active triiodothyronine (T3). DIO1 has a molecular weight around 29 kDa. This enzyme is primarily expressed in the liver kidney and thyroid gland. In these tissues DIO1 contributes to the regulation of thyroid hormone activity which affects overall metabolic rate and energy consumption at the cellular level.

Biological function summary

DIO1 serves to maintain the balance of thyroid hormones by controlling the conversion rate of T4 to T3. It functions as a monomer and does not form part of larger complexes. Its enzymatic activity ensures that cells have access to the appropriate levels of active thyroid hormone which is important for normal growth development and metabolic regulation. The expression of DIO1 and its deiodination activity are influenced by circulating thyroid hormone levels playing a feedback role in the endocrine system.

Pathways

DIO1 is a part of the thyroid hormone signaling pathway and contributes significantly to maintaining systemic hormone homeostasis. Within this pathway DIO1 interacts with other proteins such as DIO2 which also converts T4 to T3 though in different tissues and contexts. The coordinated action of these deiodinases allows precise regulation of thyroid hormone action impacting metabolic processes and developmental pathways throughout the body.

Associated diseases and disorders

DIO1 influences a range of conditions particularly thyroid-related diseases such as Graves' disease and hypothyroidism. Aberrant regulation or expression of DIO1 may disrupt the synthesis of active thyroid hormones leading to either an excess or deficit of T3. This dysregulation has connections with other proteins including thyroglobulin and thyroid-stimulating hormone receptor which are also important for proper thyroid function. Understanding DIO1's role in these disorders helps in the development of targeted therapies that aim to restore hormonal balance.
