Zinc phosphodiesterase, which displays some tRNA 3'-processing endonuclease activity (PubMed:12711671, PubMed:32075755). Specifically involved in tRNA repair: acts downstream of the ribosome-associated quality control (RQC) pathway by removing a 2',3'-cyclic phosphate from tRNAs following cleavage by ANKZF1 (PubMed:32075755). tRNAs are then processed by TRNT1 (PubMed:32075755).

Belongs to the RNase Z family.
ELAC1 also known as ElaC ribonuclease Z 1 is an enzyme involved mechanically in the processing of tRNA. It functions by endonucleolytically cleaving tRNA precursors to produce mature tRNA molecules. This protein has an approximate mass of 93 kDa and exhibits expression in various human tissues with higher levels in the liver and brain. ELAC1 acts as a zinc-dependent peptidase and shares structural similarities with the family of RNase Z enzymes.
ELAC1 plays a significant role in tRNA maturation and processing. It participates in the cleavage and removal of the 3' trailer from pre-tRNA molecules. Although not all tRNA processing events require ELAC1 its role is important where it is involved ensuring proper formation of tRNA 3' termini. ELAC1 does not form a part of larger complexes like its homolog ELAC2 which participates in mitochondrial RNA processing. It maintains its activity independently indicating specificity in its function.
ELAC1's function integrates into the broader tRNA processing pathway. This pathway is essential for proper translation and protein synthesis across eukaryotic cells. ELAC1 operates independently within these pathways but works in concert with other processing enzymes such as enzymes involved in tRNA splicing. It facilitates the maturation process that enables proper recognition and usage of tRNA by the ribosome during protein synthesis.
ELAC1 associates with certain neurological conditions due to its expression in brain tissues. Variations in ELAC1 activity or expression could potentially contribute to disorders such as Alzheimer's disease. The relationship between ELAC1 and mitochondrial proteins like ELAC2 suggests potential overlapping contributions to mitochondrial dysfunctions observed in various neurodegenerative diseases. Further research may help elucidate the exact role that ELAC1 and its interaction partners play in disease development and progression.
