EPO-R also known as erythropoietin receptor is a cell surface receptor primarily involved in erythropoiesis. The receptor has a molecular weight of around 55 kDa. It is expressed predominantly in erythroid progenitor cells and also in some non-hematopoietic tissues. IL-3RB another component of this receptor complex helps form high-affinity binding sites for erythropoietin on these cells.
Biological function summary

EPO-R operates as part of a receptor complex that is critical in promoting the survival proliferation and differentiation of erythroid progenitors. It forms a functional unit by associating with the JAK2 kinase which gets activated upon ligand binding. This activation leads to the initiation of signaling cascades that result in the production of red blood cells. IL-3RB cooperates with other cytokine receptors like GM-CSF and IL-3 adding to the complexity of cell signaling.

Pathways

This pathway is essential for transferring signals from the cell membrane to the nucleus ultimately influencing gene expression. EPO-R is associated with conditions such as anemia and polycythemia. In anemia insufficient EPO-R signaling leads to reduced red blood cell production contributing to fatigue and weakness. In polycythemia excessive activation of EPO-R can result in an over-production of red blood cells increasing the risk of blood clots. Both conditions can be assessed by investigating EPO-R interactions with proteins like JAK2 which can be mutated in such disorders.

Upon EPO stimulation, EPOR dimerizes triggering the JAK2/STAT5 signaling cascade (By similarity). In some cell types, can also activate STAT1 and STAT3 (PubMed : 11756159). May also activate the LYN tyrosine kinase (By similarity).. Isoform EPOR-T. Acts as a dominant-negative receptor of EPOR-mediated signaling.

Post-translational modifications

On EPO stimulation, phosphorylated on C-terminal tyrosine residues by JAK2 (PubMed:11781573). The phosphotyrosine motifs are also recruitment sites for several SH2-containing proteins and adapter proteins which mediate cell proliferation (PubMed:10374881). Phosphorylation on Tyr-454 is required for PTPN6 interaction, Tyr-426 for PTPN11 (By similarity). Tyr-426 is also required for SOCS3 binding, but Tyr-454/Tyr-456 motif is the preferred binding site (PubMed:12027890).. Ubiquitinated by the ECS(SOCS2) complex following ligand-binding and phosphorylation by JAK2, leading to its degradation by the proteasome (PubMed:11781573, PubMed:31182716). Regulation by the ECS(SOCS2) complex acts as a negative feedback loop of erythropoietin-mediated signaling pathway (PubMed:11781573). Ubiquitination at Lys-281 mediates receptor internalization, whereas ubiquitination at Lys-453 promotes trafficking of activated receptors to the lysosomes for degradation (By similarity). Ubiquitinated by NOSIP; appears to be either multi-monoubiquitinated or polyubiquitinated (PubMed:12746455). Ubiquitination mediates proliferation and survival of EPO-dependent cells (By similarity).
