Core histone-binding subunit that may target chromatin assembly factors, chromatin remodeling factors and histone deacetylases to their histone substrates in a manner that is regulated by nucleosomal DNA. Component of several complexes which regulate chromatin metabolism. These include the chromatin assembly factor 1 (CAF-1) complex, which is required for chromatin assembly following DNA replication and DNA repair; the core histone deacetylase (HDAC) complex, which promotes histone deacetylation and consequent transcriptional repression; the nucleosome remodeling and histone deacetylase complex (the NuRD complex), which promotes transcriptional repression by histone deacetylation and nucleosome remodeling; the PRC2 complex, which promotes repression of homeotic genes during development; and the NURF (nucleosome remodeling factor) complex.

The EZH1 EED SUZ12 RBBP4 and AEBP2 proteins form the core of Polycomb Repressive Complex 2 (PRC2) which is an essential multi-subunit protein assembly. The main function of PRC2 lies in its ability to methylate histone H3 on lysine 27 (H3K27me3) an important histone modification promoting chromatin compaction and gene silencing. The molecular weight of each subunit varies with EZH1 around 85 kDa. The PRC2 components such as EZH1 mostly express in tissues where epigenetic regulation is required including embryonic stem cells and various somatic tissues.
This complex plays a significant role in maintaining transcriptional repression across the genome contributing to cellular identity and stem cell pluripotency. PRC2 including the catalytic subunit EZH1 and its accessory proteins EED SUZ12 RBBP4 and AEBP2 facilitates chromatin remodeling and maintenance of gene silencing by regulating the histone methylation status. This biological mechanism influences differentiation pathways and is important in preserving a balance between cell proliferation and specialization.
EZH1 and its associated complex modulate important developmental and signaling pathways such as the Wnt and Notch pathways. PRC2 works closely with other chromatin modifiers and regulators like HDAC1/2 contributing to the fine-tuning of gene expression during development and cellular differentiation. These pathways are integral to the regulation of cell fate decisions and PRC2's activity ensures proper pathway signaling through chromatin-mediated control.
Abnormalities in EZH1 and associated PRC2 components link to certain cancers especially leukemia and breast cancer. Mutations or altered expression levels of PRC2 subunits influence chromatin structure and gene regulation potentially leading to oncogenesis. These components interact with other proteins such as DNMT3A and TET2 which also play roles in epigenetic modifications and cancer development. Understanding the relationship between PRC2 and disease offers potential therapeutic avenues for targeting these pathways in cancer treatment.
