The protein complex EZH2 + EED + SUZ12 + AEBP2 + RBBP4 often referred to as the Polycomb Repressive Complex 2 (PRC2) performs essential functions in chromatin remodeling. EZH2 the catalytic subunit carries out histone methylation specifically trimethylation of lysine 27 on histone H3 (H3K27me3) which leads to transcriptional repression. The full complex is a significant entity with variable mass between 200-700 kDa due to different interacting partners and post-translational modifications. PRC2's component proteins are widely expressed across different tissues playing integral roles in cellular processes.
The PRC2 complex regulates gene silencing important during development and differentiation. It forms a multiprotein complex where EZH2 works intimately with EED and SUZ12 to mediate its methyltransferase activity while AEBP2 and RBBP4 stabilize the complex and assist in DNA binding. The precise modulation of gene expression through PRC2 ensures correct lineage specification maintaining stem cell pluripotency and directing cellular differentiation.
The Polycomb Repressive Complex 2 influences major signaling networks such as the Wnt signaling and cell cycle regulation pathways. In the Wnt signaling pathway PRC2 interacts with other proteins like β-catenin to control transcription of target genes. Additionally in cell cycle regulation PRC2 modifies chromatin structures to repress genes essential for cell division when necessary. These interactions help maintain homeostasis and physiological balance in the cell.
PRC2 is linked to various cancers and developmental syndromes. Dysregulated EZH2 activity through overexpression or mutation often associates with malignancies like prostate cancer and lymphoma where it aberrantly silences tumor suppressor genes. Additionally PRC2 involvement has been noted in Weaver syndrome a developmental disorder connected with mutations in its components. The misregulation of PRC2 affects other proteins and pathways implicating it in complex disease mechanisms.
