The target EZH2 + SUZ12 + RbAp48 forms a core part of the Polycomb Repressive Complex 2 (PRC2). EZH2 also known as Enhancer of Zeste Homolog 2 is a histone methyltransferase enzyme with a mass of about 85-97 kDa depending on isoforms and post-translational modifications. SUZ12 (Suppressor of Zeste 12 protein homolog) and RbAp48 (Retinoblastoma Binding Protein 4) work with EZH2 to regulate gene expression. Together they contribute to the trimethylation of histone H3 on lysine 27 (H3K27me3) silencing target genes. The expression of this complex varies but is often found in stem cells cancer cells and tissues undergoing rapid division.
EZH2 SUZ12 and RbAp48 are significant in regulating gene expression through chromatin modification as part of the PRC2 complex. The complex plays a role in maintaining stem cell pluripotency and proper differentiation by stably repressing the transcription of specific gene sets. SUZ12 and RbAp48 stabilize the structure of the PRC2 complex facilitating the gene silencing activity of EZH2. Their activity highlights developmental processes such as cellular differentiation and proliferation.
EZH2 + SUZ12 + RbAp48 heavily influence the Wnt signaling and Notch signaling pathways. In the Wnt pathway EZH2 can repress antagonists of Wnt signals modulating cellular responses. In the Notch pathway PRC2 helps in the transcriptional repression required for cellular differentiation. Proteins such as Beta-catenin in the Wnt pathway and Notch receptors in the Notch pathway are often interlinked with the regulatory functions of this complex.
EZH2 aberrations are closely associated with cancers and developmental disorders. Overexpression or mutations in EZH2 are linked with tumorigenesis especially in prostate cancer and lymphomas. In developmental disorders mutations in SUZ12 or depletion of RbAp48 may cause syndromes characterized by intellectual disability and growth retardation. In cancers the functional connection of EZH2 with proteins like VHL (Von Hippel-Lindau tumor suppressor) can contribute to oncogenic processes by impacting pathways critical to cell cycle regulation and apoptosis suppression.
