FCN3 also known as ficolin-3 is a multifunctional protein recognized for its role in the immune system. Ficolin-3 is approximately 35 kDa in mass and is mainly expressed in the liver though it can also be found in various other tissues such as the spleen and lungs. It belongs to the ficolin family of proteins which are involved in innate immunity. The protein features a collagen-like domain and a fibrinogen-like domain facilitating its binding to sugars on microbial surfaces.
Ficolin-3 acts as an essential player in the immune response contributing to pathogen recognition and clearance. It forms a complex with other components to activate the complement pathway particularly the lectin pathway. Ficolin-3 binds to carbohydrate patterns on the surface of pathogens leading to opsonization and clearance. This activity highlights its function as a defense mechanism against infections and its role in recognizing altered cells.
Ficolin-3 facilitates the activation of the lectin complement pathway. This pathway is important for innate immune defense and involves several other proteins including mannose-binding lectin (MBL) and MASP-2 which are connected with ficolin-3 in mediating complement activation.
