Supplementary information

G protein-coupled receptor 20 variant often referred to as GPR20 belongs to the large family of G protein-coupled receptors (GPCRs). These receptors transmit signals across cell membranes upon ligand binding activating intracellular G proteins. The molecular mass of GPR20 is approximately 40 kDa. Expression of GPR20 occurs in various tissues including the brain heart and adrenal cortex. GPR20 does not have a well-known ligand making it an orphan receptor which adds complexity to its study.
Biological function summary

The GPR20 variant contributes to signal transduction processes that influence cellular responses. Researchers have not linked GPR20 to a specific ligand or described it as part of a receptor complex. It impacts cellular events through modulation of G protein activity affecting downstream signaling pathways. Although research on GPR20 is still expanding this receptor likely plays a role in normal physiological processes by influencing cellular communication.

Pathways

The signaling pathways influenced by GPR20 include the MAPK/ERK and GPCR signaling pathways. GPR20's activation can alter the signaling cascade of these pathways. Through MAPK/ERK GPR20 affects cell differentiation and proliferation while it helps mediate various cell responses in GPCR signaling. Proteins such as GRK (G protein-coupled receptor kinase) and arrestin interact with this receptor indirectly through these pathways acting as regulatory elements for signaling outcomes.

Mutations and dysregulation of GPR20 may correlate with certain cancers and cardiovascular disorders. Scientific evidence has indicated that GPR20 might associate with tumorigenesis influencing cell growth and survival in specific cancer types. It could potentially interact with proteins such as beta-arrestin in cancer progression. Additionally improper GPR20 signaling might contribute to heart-related disorders due to its expression in cardiac tissues. Consequently it creates potential for targeted therapies to mitigate these conditions.
