HIRIP3 also known as HIRA-interacting protein 3 is a protein with a mass of approximately 60 kDa. This protein is expressed in various tissues with notable presence in testis and brain. HIRIP3 mechanistically interacts with HIRA a chaperone involved in histone metabolism. The protein contains motifs that suggest a role in chromatin organization and regulation and it may be involved in transcription processes through these interactions.
Biological function summary

HIRIP3 participates in the regulation of chromatin dynamics and gene expression. It forms a complex with HIRA and other proteins linked to chromatin assembly influencing nucleosome spacing and stability. This interaction helps regulate the incorporation of histone H3.3 a variant important for active chromatin remodeling and transcriptional activation suggesting a direct role in controlling genetic activity in cells.

Pathways

Researchers have identified HIRIP3 as integral to the chromatin assembly pathway and histone modification processes. It associates with the HIRA complex impacting histone replacement during DNA repair and transcription. HIRIP3’s relationship with proteins like ASF1a indicates its involvement in chromatin remodeling processes important for maintaining genome stability and regulation of gene expression.

Disruptions in the HIRIP3 expression or function could relate to neurodegenerative diseases or infertility given its expression profile. Abnormalities in the HIRA interaction have been considered in the context of these conditions. The association of HIRIP3 with proteins involved in histone modification suggests that its dysregulation may contribute to epigenetic disorders which can lead to improper gene silencing or activation linked to pathological states.
