The spectrum was recorded with a 6545XT AdvanceBio LC/Q-TOF (Agilent Technologies) and a MabPac RP column (42.1x50 mm, 4 μm, Thermo Scientific). 5 μL of purified protein was injected and the gradient run from 85 % water : FA (99.9 : 0.1 v/v) and 15 % acetonitrile : FA (90 : 9.9 : 0.1 v/v/v) to 55 % water : FA (99.9 : 0.1 v/v) and 45 % acetonitrile : FA (90 : 9.9 : 0.1 v/v/v) within 3 minutes followed by an isocratic step for another 2.5 min. Flow rate was 0.4 mL/min and the column compartment temperature was 60 °C. Data was analysed and deconvoluted using the Bioconfirm software (Agilent Technologies).

Human IL-6 concentration was interpolated from the IL-6 standard curve. Supernatants from cell culture samples were serially diluted and assessed by the Human IL-6 ELISA kit (ab178013). Wild-type and IL-6 knockout A549 cells (ab273751) were assessed in duplicate (n=2). Cells were either treated with 20 ng/mL active recombinant human IL-1 beta protein (ab259387) for 24 h to induce expression of IL-6 or not treated with IL-1 beta. Data are represented as the mean and error bars represent standard deviation.

Flow rate was 0.5 mL/min and the column compartment temperature was 50 °C.

Initially discovered as the major endogenous pyrogen, induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production (PubMed : 3920526). Promotes Th17 differentiation of T-cells. Synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells (PubMed : 10653850). Plays a role in angiogenesis by inducing VEGF production synergistically with TNF and IL6 (PubMed : 12794819). Involved in transduction of inflammation downstream of pyroptosis : its mature form is specifically released in the extracellular milieu by passing through the gasdermin-D (GSDMD) pore (PubMed : 33377178, PubMed : 33883744). Acts as a sensor of S.pyogenes infection in skin : cleaved and activated by pyogenes SpeB protease, leading to an inflammatory response that prevents bacterial growth during invasive skin infection (PubMed : 28331908).
