Supplementary information

LACRT also known as Lacritin is a secreted glycoprotein with a molecular mass around 12-16 kDa. It is mainly expressed in the lacrimal glands and ocular surface tissues including the conjunctiva and corneal epithelium. Its presence is also identified in saliva and other secretions. As a multifunctional protein LACRT plays an important role in maintaining eye health especially in tear film stability and ocular surface homeostasis.
Biological function summary

LACRT contributes to cell proliferation survival and hydration of epithelial cells. It is not part of a complex but interacts with various cellular pathways to exert its functions. LACRT promotes the secretion of tears and facilitates the binding of tears to the ocular surface which are key aspects in sustaining the health of the eye’s surface. Also it has antimicrobial properties providing a protective barrier against potential pathogens.

Pathways

LACRT participates in pathways like the MAPK and Wnt signaling pathways where it influences cellular growth and differentiation. Through these pathways LACRT interacts with proteins such as ERK1/2 and β-catenin which are important for its regulatory functions in the eye. The protein modulates signaling processes that maintain tissue integrity and repair.

LACRT shows relevance to conditions like dry eye disease and Sjögren's syndrome. In these disorders LACRT levels often decrease leading to impaired tear secretion and uncomfortable ocular symptoms. This protein potentially links to other proteins such as aquaporins which play roles in fluid transport and ocular moisture maintenance impacting the severity of these conditions.
