Leucine-Rich Repeat Transmembrane Neuronal Protein 2 (LRRTM2) also known as C22orf34 is a protein with a molecular weight of approximately 68 kDa. This protein plays a role in synapse formation by promoting synaptogenesis and maintaining synaptic stability. LRRTM2 is highly expressed in the central nervous system especially in the hippocampus and cortex regions of the brain. It also locates primarily at the postsynaptic membranes of excitatory synapses where it interacts with extracellular matrix components and other cell adhesion molecules.
Biological function summary

LRRTM2 is essential for modulating excitatory synaptic strength and plasticity. The protein is part of a larger complex interacting with presynaptic neurexins which are involved in synapse specification. These interactions help stabilize the synapse and influence synaptic remodeling. LRRTM2 contributes to the efficient transmission of nerve signals by regulating the recruitment of presynaptic vesicles and postsynaptic receptors shaping the communication network within the neuronal circuits.

Pathways

LRRTM2 is a component of synaptic transmission and plasticity pathways. It plays a role in the synaptogenesis pathway involving neurexins and neuroligins which are important for synapse development and maturation. The balance and interaction between LRRTM family members like LRRTM1 and LRRTM3 influence synaptic connections and can modulate neural circuit assembly. LRRTM2's function within these pathways impacts the strength and specificity of synaptic interactions essential for learning and memory processes.

LRRTM2 has associations with psychiatric conditions such as schizophrenia and autism spectrum disorder. LRRTM2's disruption can impair synaptic plasticity leading to altered neural connectivity observed in these disorders. Changes in the expression or function of proteins like neurexins which interact with LRRTM2 can further contribute to disease pathologies. Understanding LRRTM2's role in these contexts could help identify therapeutic targets for modifying synaptic connectivity affected in such conditions.
