Belongs to the immunoglobulin superfamily. IgLON family.
The limbic system-associated membrane protein (LSAMP) also known as IgLON5 is a protein with a mass of around 64 kDa. It is an important component of the neuronal membrane and found heavily expressed in the limbic system which includes areas like the hippocampus and amygdala. LSAMP localizes at synaptic junctions suggesting its active role in neuron-to-neuron communication. It belongs to the immunoglobulin superfamily and is known for facilitating cell adhesion processes in the brain.
LSAMP influences the growth and connectivity of neurons. It plays an important role in neuronal development guiding axonal processes and stabilizing synaptic contacts. LSAMP does not function alone; it forms part of a complex with several other cell adhesion molecules working collaboratively to influence neural circuit formation and synaptic plasticity. These actions are important for the correct wiring of nerve cells during development and learning processes.
LSAMP influences the cellular signaling related to growth and connectivity. It is active in pathways such as the MAPK signaling pathway which affects cell division survival and differentiation. LSAMP interacts with proteins like NCAM (neural cell adhesion molecule) and L1CAM (L1 cell adhesion molecule) which are significant in these pathways. These proteins work together to modulate molecular mechanisms that underlie neural network stability and response to environmental stimuli.
LSAMP has been connected to abnormalities in neural circuits leading to potential links with psychiatric disorders such as schizophrenia and anxiety. Research indicates that alterations in LSAMP expression or function can disrupt neural development and contribute to these disorders. It may act in conjunction with factors like BDNF (brain-derived neurotrophic factor) during disease pathogenesis highlighting the complex interactions that underpin these neurosignificant disorders.
