Function

Component of the Mediator complex, a coactivator involved in the regulated transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II and the general transcription factors.

Activity summary

The MED3 protein also known as Vitamin D3 receptor interacting protein complex 12 (DRIP12) possesses a mass of approximately 64 kDa. This protein acts as part of the larger Mediator complex which plays an important role in the regulation of gene transcription. MED3 is ubiquitously expressed in various tissues reflecting its essential function in gene expression. It modulates transcription by serving as a bridge between transcription factors and RNA polymerase II.

Biological function summary

Functions of MED3 extend to the facilitation of transcriptional activation processes. Being a part of the Mediator complex MED3 influences RNA polymerase II-dependent transcription in eukaryotic cells. It works closely with other core subunits to recruit the complex to gene promoters and enable efficient transcription initiation. MED3 by interacting with transcriptional regulators adjusts the transcriptional output necessary for cell growth and differentiation.

Pathways

Studies show that MED3 is involved in the regulation of Wnt signaling and the Notch signaling pathways. These pathways are key in cell fate determination and developmental processes. The MED3 protein intersects with the activities of other transcriptional regulators like beta-catenin in the Wnt pathway emphasizing its impact on important developmental gene expression.

Associated diseases and disorders

MED3 holds an association with certain cancers and developmental disorders. Dysregulation of MED3 activity may contribute to colorectal cancer implicating its involvement with the aberrant activation of Wnt signaling. In some developmental disorders the improper function of MED3 can disrupt normal transcriptional regulation also impacting Notch signaling-related proteins. Understanding MED3's role in these contexts provides insights into its potential as a therapeutic target.
