Chaperone specifically assisting the folding of beta-propeller/EGF modules within the family of low-density lipoprotein receptors (LDLRs) (PubMed:15014448). Acts as a modulator of the Wnt pathway through chaperoning the coreceptors of the canonical Wnt pathway, LRP5 and LRP6, to the plasma membrane (PubMed:17488095). Essential for specification of embryonic polarity and mesoderm induction. Plays an essential role in neuromuscular junction (NMJ) formation by promoting cell-surface expression of LRP4 (By similarity). May regulate phagocytosis of apoptotic retinal pigment epithelium (RPE) cells (By similarity).

MESDC2 also known as Mesoderm Development Candidate 2 is an endoplasmic reticulum (ER) resident chaperone protein. It exhibits a molecular weight of approximately 28 kDa. This protein is highly expressed in tissues where receptor maturation processes are significant. MESDC2 plays an important role in the folding and transport of low-density lipoprotein receptors (LDLRs) to the cell surface important for their proper function. Besides the ER it exhibits expression patterns in developing mesodermal tissues.
The function of MESDC2 involves regulation of cholesterol homeostasis and cellular lipid uptake processes. MESDC2 acts as part of the molecular machinery that ensures the proper folding of specific receptor proteins within the ER especially the LDL receptor family. It is critical for receptor maturation before they exit the ER and proceed to the Golgi. MESDC2 therefore indirectly influences membrane-bound receptor availability impacting signaling pathways that depend on these receptors.
MESDC2 is primarily involved in lipid metabolism and transport pathways. It facilitates the LDL receptor's correct folding impacting cholesterol uptake and lipid homeostasis. These pathways are interconnected with proteins such as PCSK9 which modulates the recycling and degradation of LDL receptors. MESDC2's role supports efficient cholesterol transport preventing disruptions that might otherwise lead to cellular and systemic imbalances.
MESDC2 plays a role in conditions linked with cholesterol and lipid metabolism. Aberrations in its function have associations with cardiovascular diseases like atherosclerosis. The protein's interactions with LDL receptors connect it to hypercholesterolemia where dysfunctional receptor activity leads to elevated cholesterol levels. MESDC2's role in these processes further links it to the broader regulatory network including PCSK9 and its impact on lipid level modulation.
