Supplementary information

MKX also known as Mohawk Homeobox is a transcription factor with a mass of approximately 36 kDa. This protein plays a role in gene regulation by binding to DNA sequences and influencing the expression of target genes. MKX mainly expresses in mesodermal tissues especially in skeletal muscles tendons and ligaments during development. Its expression is spatially and temporally regulated reflecting its involvement in tissue-specific processes.
Biological function summary

MKX influences the formation and maintenance of fibrous connective tissues. It does not function as part of a known complex but interacts with other transcription factors to regulate gene expression patterns essential for tissue differentiation and function. MKX is involved in orchestrating the development of musculoskeletal structures by modulating the expression of genes related to extracellular matrix composition and organization. Through these actions MKX contributes to the structural integrity and mechanical properties of tendons and ligaments.

Pathways

MKX plays a role in the tendon development and TGF-β signaling pathways. In these pathways MKX coordinates with proteins such as Scleraxis (SCX) and Tenomodulin (TNMD) to promote tendon and ligament differentiation. The interaction of MKX within these pathways facilitates cellular processes that mediate tissue formation and repair indicating its integral role in connective tissue physiology.

MKX has strong connections to conditions involving tendon and ligament degeneration such as tendinopathy and osteoarthritis. Altered expression levels or mutations in MKX may disrupt normal tissue development and repair processes linking it to these disorders. Furthermore MKX interacts with proteins like Matrix Metalloproteinases (MMPs) which play a role in tissue remodeling and are involved in pathological conditions when dysregulated. Understanding the functions and interactions of MKX aids in exploring therapeutic targets for musculoskeletal diseases.
