NECAP2 also known as NECAP endocytosis associated 2 is a protein that plays an important role in endocytic processes. Structurally it weighs approximately 27 kDa. This protein is highly expressed in the brain indicating its potential importance in neural activities. NECAP2 contains a phosphoinositide-binding PH domain and a C-terminal DPF motif which help it interact with other endocytic proteins.
NECAP2 contributes to the efficiency of clathrin-mediated endocytosis. It acts as an accessory protein involved in the formation of clathrin-coated pits and vesicles. Part of the endocytic machinery NECAP2 works with AP-2 adaptor complex a significant player in endocytosis. Its interaction with clathrin and other adaptor proteins suggests that NECAP2's function is essential for regulating receptor-mediated internalization of molecules.
NECAP2 participates in the clathrin-mediated endocytic pathway which is important for cellular nutrient uptake receptor recycling and signal transduction. NECAP2 closely interacts with proteins like clathrin and AP-2 helping modulate vesicle formation. In addition to its role in endocytosis NECAP2 might influence other cellular processes like signal transduction by affecting how specific receptors are recycled on cell membranes.
NECAP2 has associations with neurological disorders due to its high expression in the brain and involvement in brain-specific endocytic pathways. Alterations in its normal function can relate to conditions like Alzheimer’s disease where endocytic processing of amyloid precursor protein is important. Moreover NECAP2's dysregulation could also influence schizophrenia through its interactions with proteins involved in synaptic vesicle endocytosis and neurotransmitter release.
