Involved in base excision repair of DNA damaged by oxidation or by mutagenic agents. Has DNA glycosylase activity towards 5-hydroxyuracil and other oxidized derivatives of cytosine with a preference for mismatched double-stranded DNA (DNA bubbles). Has low or no DNA glycosylase activity towards thymine glycol, 2-hydroxyadenine, hypoxanthine and 8-oxoguanine. Has AP (apurinic/apyrimidinic) lyase activity and introduces nicks in the DNA strand. Cleaves the DNA backbone by beta-delta elimination to generate a single-strand break at the site of the removed base with both 3'- and 5'-phosphates.
Endonuclease 8-like 2, DNA glycosylase/AP lyase Neil2, DNA-(apurinic or apyrimidinic site) lyase Neil2, Endonuclease VIII-like 2, Nei homolog 2, Nei-like protein 2, NEH2, NEIL2

NEIL2 also known as Nei-like 2 is a DNA glycosylase involved in base excision repair important for maintaining genomic integrity. NEIL2 is a protein with a mass of approximately 38 kDa. This protein mainly finds expression in actively dividing cells including various tissues like the brain liver and lungs. NEIL2 acts by recognizing oxidative DNA damage and excising the damaged base which initiates the repair process.
NEIL2 participates in the vital process of repairing oxidative DNA lesions. It possesses a strong affinity for removing oxidized pyrimidines which helps prevent genomic instability. NEIL2 operates as part of the DNA repair machinery sometimes interacting with other proteins in complexes to facilitate repair processes. Its activity ensures the accurate restoration of proper DNA structure contributing to cellular health and survival.
NEIL2 functions in the base excision repair pathway important for rectifying oxidative DNA damage. This pathway includes NEIL2's interaction with important proteins like APE1 which further processes the DNA strand breaks generated by NEIL2. Another pathway associated with NEIL2 is the nucleotide excision repair pathway. This linkage suggests its potential role in repairing more complex DNA lesions connecting it with key repair proteins.
NEIL2 shows association with cancer and neurodegenerative diseases. The protein's ability to maintain genomic stability plays a role in cancer prevention as malfunction can lead to accumulation of DNA mutations driving tumorigenesis. Additionally defective NEIL2 function affects neuronal cells linking it to disorders like Parkinson's disease. Its interactions with proteins such as OGG1 highlight its involvement in defending against oxidative stress which is a factor in various diseases.
