Function

Phosphorylates serines and threonines, but also appears to possess tyrosine kinase activity (PubMed:20230784). Involved in DNA damage checkpoint control and for proper DNA damage repair (PubMed:20230784). In response to injury that includes DNA damage, NEK1 phosphorylates VDAC1 to limit mitochondrial cell death (PubMed:20230784). May be implicated in the control of meiosis (By similarity). Involved in cilium assembly (PubMed:21211617).

Supplementary info

Activity summary

NEK1 also known as NIMA-related kinase 1 plays an important role in cell cycle regulation and DNA damage response. This serine/threonine-protein kinase has a molecular mass of approximately 140 kDa. NEK1 shows expression across various tissues particularly in the brain and testis. It functions by phosphorylating other proteins modifying their activity and ensuring proper cell division and stress response.

Biological function summary

NEK1 coordinates critical processes such as ciliogenesis and mitochondrial function. It participates as part of protein complexes that help maintain cellular integrity. NEK1 interacts with proteins to regulate centrosome integrity and stability ensuring proper cell polarization and division. Additionally NEK1 has a role in the DNA damage checkpoint a mechanism that maintains genomic stability.

Pathways

NEK1 integrates into the DNA damage response pathway and cilia-related signaling pathways. It interacts with key proteins such as ATR and ATM to mediate cell cycle arrest after DNA damage. NEK1 also influences the Hedgehog signaling pathway being important for ciliary function. These interactions underline its importance in cell cycle control and signaling.

Associated diseases and disorders

NEK1 associates closely with amyotrophic lateral sclerosis (ALS) and polycystic kidney disease (PKD). Mutations in NEK1 have been linked to familial ALS indicating its role in neurodegenerative processes. In PKD NEK1 interacts with the polycystin proteins which are essential for kidney function leading to cyst formation when disrupted. These connections illustrate the impact of NEK1 dysregulation on multiple disease states.
