NEK4 also known as NimA-related protein kinase 4 functions as a serine/threonine kinase involved in cell cycle regulation and ciliogenesis. It has a molecular weight of about 84 kDa. NEK4 is expressed widely in various tissues with significant expression in lung skeletal muscle and kidney. Its expression is important for proper cellular processes like mitosis and maintenance of the ciliary structure.
NEK4 participates in centrosome function and cilia dynamics playing an important role in maintaining cellular integrity. It associates with other proteins in a complex manner to facilitate cell cycle progression. NEK4 shows importance in regulating the stability of microtubules which affects the proper formation and function of cilia.
NEK4 integrates into major cellular pathways involved in cell cycle checkpoints and ciliogenesis. It is a part of the cell cycle regulation pathway influencing events necessary for DNA replication and mitotic progression. NEK4 also interacts with proteins like NEK1 which further modulates its activity in regulating cellular responses to DNA damage and ensuring correct cell division.
NEK4 links to conditions such as cancer and primary ciliary dyskinesia. In various cancers NEK4 expression may influence tumor growth and metastasis through its role in cell cycle regulation. In primary ciliary dyskinesia faults in NEK4-related proteins like NEK1 can affect cilia structure and function leading to clinical symptoms.
