The NLRP3 protein also known as NACHT LRR and PYD domains-containing protein 3 and NEK7 known as NIMA-related kinase 7 play integral parts in cellular pathways. NLRP3 has a molecular mass of approximately 118 kDa and is expressed mainly in innate immune cells such as macrophages and dendritic cells. NEK7 with a mass around 34 kDa is largely expressed in dividing cells and performs important cellular roles during the cell cycle. These proteins are part of the NLRP3 inflammasome complex a critical component of the immune response which detects pathogenic microorganisms and stress signals.
Biological function summary

NLRP3 and NEK7 are involved in innate immunity playing key roles in inflammation. The NLRP3 inflammasome comprises NLRP3 ASC (apoptosis-associated speck-like protein containing a CARD) and pro-caspase-1. NEK7 acts as a positive regulator of NLRP3 inflammasome activation. This complex once formed leads to the activation of caspase-1 which then processes pro-inflammatory cytokines like IL-1β and IL-18 into their active forms. This process aids in the defense against infections but its dysregulation can result in excessive inflammation.

Pathways

NLRP3 and NEK7 are instrumental in the inflammasome and IL-1 signaling pathways. Within the inflammasome signaling NLRP3 acts as a pattern recognition receptor and its interaction with NEK7 is critical for sensing cell danger signals. This recognition and activation also involve other proteins such as ASC linking the inflammasome pathway to wider immune responses. The IL-1 signaling pathway further connects NLRP3 inflammasome activity to downstream effects promoting inflammatory responses and influencing adaptive immunity.

NLRP3 and NEK7 are linked to autoinflammatory diseases and neurodegenerative disorders. One well-studied association is with Cryopyrin-associated periodic syndromes (CAPS) where mutations in NLRP3 lead to abnormal inflammasome activation causing chronic inflammation. Another connection is identified in Alzheimer’s disease where the pathologic activation of the NLRP3 inflammasome contributes to neuroinflammation.
