Supplementary information

NSL1 also known as "Novel Spindle" or "Nucleosome Assembly Protein 1-Like 1" plays a mechanical role in the stabilization of centromeres during cell division. The NSL1 protein has a molecular mass of approximately 28 kDa. It primarily expresses in tissues with high proliferative indices such as bone marrow and the gastrointestinal tract. Due to its localisation during mitosis NSL1 is vital for proper chromosome segregation.
Biological function summary

NSL1 is a component of the Mis12 complex which associates with the kinetochore in eukaryotic cells. This protein complex ensures the accurate attachment of chromosomes to the mitotic spindle microtubules. NSL1’s interaction with other members of the Mis12 complex is key in establishing proper kinetochore-microtubule connections. Consequently it contributes significantly to the fidelity of chromosome segregation and stability.

Pathways

NSL1's function is critical in the chromosome segregation pathway and cell cycle regulation. Within these pathways NSL1 interacts with proteins such as CENP-E and MAD2 which help monitor and correct erroneous microtubule attachments. Both pathways ensure accurate division of chromosomes preventing aneuploidy and its associated consequences. NSL1's role highlights its importance in maintaining genomic integrity during cell division.

Aberrant NSL1 function has a connection with cancer and chromosomal instability syndromes. In cancer mutations or altered expression levels of NSL1 might disrupt the normal functioning of the Mis12 complex impacting proteins like BubR1 and Aurora B kinase. These disruptions could lead to improper chromosome segregation and tumorigenesis. Understanding NSL1’s involvement can provide insights into therapeutic targets for related diseases.
