Component of the cytosolic iron-sulfur (Fe/S) protein assembly (CIA) machinery. Required for maturation of extramitochondrial Fe-S proteins. The NUBP1-NUBP2 heterotetramer forms a Fe-S scaffold complex, mediating the de novo assembly of an Fe-S cluster and its transfer to target apoproteins. Negatively regulates cilium formation and structure.
Cytosolic Fe-S cluster assembly factor NUBP2, Nucleotide-binding protein 2, NBP 2, NUBP2

Belongs to the Mrp/NBP35 ATP-binding proteins family. NUBP2/CFD1 subfamily.
NUBP2 also known as nucleotide-binding protein 2 functions as an ATPase and plays a role in cellular iron-sulfur cluster assembly. This protein has a mass of approximately 32 kDa. NUBP2 localizes in both the cytoplasm and nucleus indicating a dynamic cellular distribution. Its expression occurs in various tissues but it shows significant levels in highly proliferative tissues suggesting a link to cellular growth and metabolism.
NUBP2 associates with the cytosolic iron-sulfur assembly (CIA) complex. This complex is essential for the maturation of iron-sulfur clusters which are important for the function of a wide range of enzymes. By contributing to this maturation NUBP2 supports enzymatic activities that are necessary for the stability and function of proteins involved in DNA synthesis and repair as well as mitochondrial energy metabolism.
NUBP2 participates in the cytosolic iron-sulfur protein assembly pathway. This pathway is critical for biogenesis of iron-sulfur clusters used in various cellular processes. NUBP2 functions alongside other CIA components such as NUBP1 and CIAPIN1. By facilitating the transfer of iron-sulfur clusters to target apoproteins NUBP2 ensures the proper function of enzymes involved in processes like DNA repair and metabolic regulation.
Disruptions in NUBP2 function link to certain types of cancer and neurological disorders. Aberrant expression or mutations in NUBP2 may contribute to tumorigenesis as the proteins reliant on iron-sulfur clusters play pivotal roles in cell cycle regulation. Additionally defects in NUBP2 and its interacting partners such as NUBP1 may result in neurological impairments because of the important role of iron-sulfur clusters in neuronal function and maintenance.
