Biological function summary
The function of ORAV1 extends beyond being a mere channel. It forms part of a larger complex known as the calcium release-activated calcium channel modulator (CRAC) complex. A critical partner for ORAV1 in this complex is the stromal interaction molecule 1 (STIM1) which senses depletion of calcium in the endoplasmic reticulum. The interaction between ORAV1 and STIM1 triggers calcium influx across the cell membrane which is fundamental for the activation of various cellular signaling cascades.

ORAV1 plays a central role in calcium signaling pathways. It is involved in pathways such as the immune response pathway where calcium ions function as secondary messengers. The interaction between ORAV1 and STIM1 regulates the activation of the nuclear factor of activated T-cells (NFAT) pathway which is essential for gene expression in lymphocytes. This calcium-dependent signaling is critical for proper immune function and cellular metabolism regulation.

The complex LTO1 : YAE1 functions as a target specific adapter that probably recruits apo-ABCE1 to the cytosolic iron-sulfur protein assembly (CIA) complex machinery (PubMed : 26182403). May be required for biogenesis of the large ribosomal subunit and initiation of translation (PubMed : 23318452). May play a role in the regulation of proline metabolism and ROS production (PubMed : 24930674).
