Participates in the apoptotic response to DNA damage. Isoforms containing the transactivation domain are pro-apoptotic, isoforms lacking the domain are anti-apoptotic and block the function of p53 and transactivating p73 isoforms. May be a tumor suppressor protein. Is an activator of FOXJ1 expression (By similarity). It is an essential factor for the positive regulation of lung ciliated cell differentiation (PubMed:34077761).

Belongs to the p53 family.
Isoform alpha (but not isoform beta) is sumoylated on Lys-627, which potentiates proteasomal degradation but does not affect transcriptional activity. Phosphorylation by PLK1 and PLK3 inhibits the transcription regulator activity and pro-apoptotic function.
The protein p73 beta also known as TP73 mechanically functions as a transcription factor encoded by the TP73 gene. It has a mass of approximately 70 kDa. P73 beta is expressed in various tissues with a significant presence in the brain heart and skeletal muscle. This protein shares structural similarity with the tumor suppressor p53 and both belong to the same protein family. P73 beta plays an important role in gene expression regulation particularly in contexts related to cellular responses to stress and DNA damage.
P73 beta regulates cell cycle and apoptosis. It participates in the p53 family protein complex allowing it to engage in cellular processes that control cell growth and programmed cell death. The protein maintains genomic stability and contributes to the cellular response to diverse stress signals. By activating target genes involved in apoptosis p73 beta enhances cellular defense mechanisms against harmful conditions.
The protein p73 beta takes part in the ATM/ATR signaling and p53 pathways both central in cellular responses to DNA damage. Within these pathways p73 beta modulates downstream signaling that determines cell cycle arrest or apoptosis. Its interaction with other proteins such as p53 and p63 illustrates the interconnected regulation within these pathways. P73 beta can influence outcomes in cells based on its regulatory network with these proteins coordinating precise cellular responses to stressors.
Mutations or dysregulation of p73 beta have correlations with various cancers including neuroblastoma and breast cancer. The protein's role in tumor suppression and cellular regulation makes it significant in cancer research. Associations with proteins like p53 emphasize its involvement in maintaining normal cellular functions and preventing malignant transformation. Studies continue to investigate how alterations in p73 beta expression or function contribute to oncogenesis and cancer progression highlighting its potential as a therapeutic target.
