Biological function summary
PARP11 modifies proteins by adding ADP-ribose units impacting their interaction and activity. It is not part of a large complex but works on its own. Its activity influences cellular responses to stress and can alter how cells communicate and survive under adverse conditions. These functions highlight its significance in controlling immune responses and inflammation.
Pathways
PARP11 contributes to the regulation of the NF-kB signaling and interferon pathways. These pathways are essential for immune function and inflammation control. PARP11 interacts with other proteins like PARP1 in the NF-kB pathway impacting transcriptional activity and inflammatory responses. Its actions help modulate cellular responses to DNA damage and stress signals.

General info
Function
Mono-ADP-ribosyltransferase that mediates mono-ADP-ribosylation of target proteins (PubMed : 25043379, PubMed : 25673562). Plays a role in nuclear envelope stability and nuclear remodeling during spermiogenesis (By similarity). Inhibits the type I interferon activated signaling pathway (PubMed : 30988430). Mechanistically, mono-ADP-ribosylates beta-TrCP/BTRC to promote IFNAR1 ubiquitination and protect BTRC from ubiquitin-proteasome degradation. Additionally, acts as an antiviral factor by cooperating with PARP12 to suppress Zika virus replication, independent of IFNAR1 regulation or intrinsic PARP enzymatic activity (PubMed : 34187568). Instead, facilitates the degradation of viral NS1 and NS3 proteins, potentially disrupting viral replication (PubMed : 34187568).
Sequence similarities
Belongs to the ARTD/PARP family.
Post-translational modifications
Auto-mono-ADP-ribosylated.
