Potential calcium-dependent cell-adhesion protein.
PCDH10 known also as Protocadherin-10 is a member of the cadherin superfamily responsible for calcium-dependent cell-cell adhesion. This protein typically has a mass of around 130 kDa and is characterized by its large extracellular domain composed of cadherin repeats. PCDH10 is mainly expressed in the brain particularly in regions involved in learning and memory. Its expression patterns suggest that it plays a significant role in neural development and synaptic connectivity.
Protocadherin-10 is involved in the establishment and maintenance of specific cell-cell connections. It is not part of any known complex but interacts with cytoskeletal elements to facilitate cellular architecture and signaling. PCDH10 thereby contributes to proper functioning of neuronal circuits. The correct expression of PCDH10 influences the dynamics of neuronal networks and neuronal communication.
PCDH10 participates in signaling pathways linked to neuronal development and synaptic plasticity such as the Wnt pathway. It is indirectly related to several proteins within the Wnt pathway that are important in guiding neuronal growth and differentiation. PCDH10 may work alongside other protocadherins interacting with signaling cascades that coordinate synapse formation and stabilization.
Several studies have implicated abnormalities in PCDH10 expression with neuropsychiatric disorders including autism spectrum disorder (ASD) and schizophrenia. These disorders often show disrupted neural connectivity likely linked to altered PCDH10 function. In ASD for instance abnormalities in PCDH10 potentially interact with other proteins such as neuroligins that play roles in synaptic formation and function hinting at complex interplay in these pathologies.
