Biological function summary
The PCMTD1 enzyme prevents the accumulation of damaged proteins which could otherwise disrupt cellular functions. It forms part of a greater protein repair mechanism working alongside other correction processes to maintain protein integrity. Specifically PCMTD1 contributes to maintaining protein stability and functionality preventing erroneous protein folding that might disrupt normal cellular operations.
Pathways
Researchers place PCMTD1 within the protein repair and degradation pathways linking it to cellular proteostasis. It interacts with other enzymatic components involved in protein turnover such as the ubiquitin-proteasome system. This collaboration ensures damaged proteins are repaired or removed to uphold cell homeostasis. PCMTD1's role in these pathways is important for maintaining the balance between protein synthesis and degradation which is necessary for normal cellular functions.

General info
Function
Substrate recognition component of an ECS (Elongin BC-CUL5-SOCS-box protein) E3 ubiquitin ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins (PubMed : 35486881). Specifically binds to the methyltransferase cofactor S-adenosylmethionine (AdoMet) via the N-terminal AdoMet binding motif, but does not display methyltransferase activity (PubMed : 35486881). May provide an alternate maintenance pathway for modified proteins by acting as a damage-specific E3 ubiquitin ligase adaptor protein (PubMed : 35486881).
Sequence similarities
Belongs to the methyltransferase superfamily. L-isoaspartyl/D-aspartyl protein methyltransferase family.
