Assayed using a PDE4A Assay Kit. Assay conditions: 10 mM Tris-HCl, pH 7.4, 10 mM MgCl2, 0.1 mg/ml BSA, 0.05% Tween-20, 200 μM cAMP, 2.5 kU 5'-nucleotidase, and serial dilutions of ab198423 at 37°C for 20 min. Quantified by 5'- nucleotidase cleaving the 5' -AMP product and releasing the phosphate group which is detected by Malachite Green Reagent.

PDE4A or phosphodiesterase 4A is an isoform part of the PDE4 family known for hydrolyzing cAMP an important second messenger in cells. The mass of PDE4A can vary depending on the isoform with commonly referenced masses around 70-100 kDa. This enzyme is expressed in various tissues including the brain immune cells and lungs. Other popular names for this protein include PDE46 and DPDE2 indicating its different isoforms and specific functionalities across tissue types.

Biological function summary

PDE4A plays an important role in regulating intracellular levels of cyclic adenosine monophosphate (cAMP) a critical component in signaling pathways that affect numerous cellular processes. By controlling cAMP concentrations PDE4A regulates inflammatory responses and airway smooth muscle contraction which is important for respiratory functions.
