Unit Definition: One unit is defined as the amount of enzyme that will convert 1 pmole of 3', 5'-cAMP to 5'-AMP per minute at 37°C.
Assayed using a PDE4A Assay Kit. Assay conditions: 10 mM Tris-HCl, pH 7.4, 10 mM MgCl2, 0.1 mg/ml BSA, 0.05% Tween-20, 200 μM cAMP, 2.5 kU 5'-nucleotidase, and serial dilutions of ab198423 at 37°C for 20 min. Quantified by 5'- nucleotidase cleaving the 5' -AMP product and releasing the phosphate group which is detected by Malachite Green Reagent.

PDE4A or phosphodiesterase 4A is an isoform part of the PDE4 family known for hydrolyzing cAMP an important second messenger in cells. The mass of PDE4A can vary depending on the isoform with commonly referenced masses around 70-100 kDa. This enzyme is expressed in various tissues including the brain immune cells and lungs. Other popular names for this protein include PDE46 and DPDE2 indicating its different isoforms and specific functionalities across tissue types.
PDE4A plays an important role in regulating intracellular levels of cyclic adenosine monophosphate (cAMP) a critical component in signaling pathways that affect numerous cellular processes. By controlling cAMP concentrations PDE4A regulates inflammatory responses and airway smooth muscle contraction which is important for respiratory functions. PDE4A does not function within a complex but operates as a homodimer when exerting its effects on signaling cascades.
PDE4A's role is significant in the cAMP-dependent signaling pathway. This pathway influences processes like inflammation and smooth muscle relaxation. PDE4A interacts with various proteins such as PKA (protein kinase A) which is activated by cAMP and mediates several downstream effects. By influencing the cAMP pathway PDE4A affects bodily responses including immune and respiratory systems.
PDE4A has implications in conditions such as asthma and chronic obstructive pulmonary disease (COPD). Dysregulation of cAMP through PDE4A can contribute to these respiratory conditions by affecting airway inflammation and muscle tone. PDE4A is also linked to other related proteins such as PDE4B which can also impact inflammatory pathways in similar disease contexts. PDE4 inhibitors have been explored as therapeutic agents for modifying these diseases by targeting the enzymatic activity of PDE4A and related isoforms to restore balanced cAMP levels.
