The protein phosphatase methylesterase 1 (PME-1) also known as protein phosphatase 2A methylesterase is an enzyme that regulates protein function by demethylation. PME-1 has a mass of approximately 42 kDa. It primarily resides in the cytoplasm and nucleus of cells with high expression levels in brain tissue. Its function involves the removal of methyl groups from the carboxyl-terminal leucine residue of the catalytic subunit of protein phosphatase 2A (PP2A). This action regulates the activity states of PP2A a major serine/threonine phosphatase.

PME-1 forms part of the regulatory complexes of PP2A modulating its methylation state and associated functional properties.
