The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion. That Rab is involved in transcytosis, the directed movement of endocytosed material through the cell and its exocytosis from the plasma membrane at the opposite side. Mainly observed in epithelial cells, transcytosis mediates for instance, the transcellular transport of immunoglobulins from the basolateral surface to the apical surface. Most probably controls membrane trafficking through apical recycling endosomes in a post-endocytic step of transcytosis. Required for melanosome transport and release from melanocytes, it also regulates dendrite and dendritic spine development (By similarity). May also play a role in cell migration.

Rab17 also known by its other name RAB17 member RAS oncogene family is a small GTPase with a molecular mass of approximately 25 kDa. It participates in the regulation of membrane trafficking particularly during transcytosis. Rab17 is mainly expressed in epithelial cells. The presence of Rab17 is most notable in polarized cells where its function is closely associated with the sorting and transport of molecules across distinct membrane domains.
Rab17 influences intracellular transport processes within cells. It associates with recycling endosomes and is integral in epithelial cell transcytosis which is the process that allows the transport of material from one side of a cell to the other. Rab17 does not function in isolation. It often forms complexes with proteins like Rab11 playing an important role in maintaining cell polarity and facilitating efficient vesicle-mediated transport within polarized epithelial cells.
Rab17 plays a significant role in the transcytotic pathways present in epithelial cells which is vital for maintaining differentiated cellular functions. It works closely with the recycling pathway facilitated by Rab11 ensuring proteins and lipids are correctly sorted and transported to the appropriate domain of the plasma membrane. Rab17 is part of the wider Rab protein family which collectively regulates various intracellular vesicle trafficking routes essential for cellular homeostasis.
Dysfunctions in Rab17 have been linked to renal disorders particularly affecting proximal tubular functions due to its expression in kidney epithelial cells. Abnormalities in Rab17-mediated trafficking processes can disrupt the biochemical balance within cells impacting renal reabsorption processes. Additionally Rab17 has connections to epithelial-related cancers where altered expression or mutations can result in dysregulated cell polarity and contribute to tumorigenesis. Its interplay with proteins like Rab11 can influence cancer progression by affecting cell adhesion and migration pathways.
